چکیده

Remarkable methodological advances in the past decade have expanded the application of liquid chromatography coupled with mass spectrometry (LC/MS) analysis of biotherapeutics. Currently, LC/MS represents a promising alternative or supplement to the traditional ligand binding assay (LBA) in the pharmacokinetic, pharmacodynamic, and toxicokinetic studies of protein drugs, owing to the rapid and cost-effective method development, high specificity and reproducibility, low sample consumption, the capacity of analyzing multiple targets in one analysis, and the fact that a validated method can be readily adapted across various matrices and species. While promising, technical challenges associated with sensitivity, sample preparation, method development, and quantitative accuracy need to be addressed to enable full utilization of LC/MS. This article introduces the rationale and technical challenges of LC/MS techniques in biotherapeutics analysis and summarizes recently developed strategies to alleviate these challenges. Applications of LC/MS techniques on quantification and characterization of antibody biotherapeutics are also discussed. We speculate that despite the highly attractive features of LC/MS, it will not fully replace traditional assays such as LBA in the foreseeable future; instead, the forthcoming trend is likely the conjunction of biochemical techniques with versatile LC/MS approaches to achieve accurate, sensitive, and unbiased characterization of biotherapeutics in highly complex pharmaceutical/biologic matrices. Such combinations will constitute powerful tools to tackle the challenges posed by the rapidly growing needs for biotherapeutics

Epidemic Intermittent Fever in the Mauritius

fever had been registered in the Military Hospitals between the years 1823 and 1858. In the Civil Hospital for four years prior to 1866, (including all classes of the community,) only 85 cases of paludal fever had occurred j whereas in the same hospitals during the eighteen months ending June 1867, no less than 2,576 patients were under treatment suffering from intermittent fever ; and within t...

Dr Thomas Hodgkin, Dr Martin Delany, and the "return to Africa".

The medical accomplishments of Thomas Hodgkin (1798-1866) are well known. The introduction of the stethoscope and creation of the pathological museum at Guy's Hospital, London, his lectures on morbid anatomy, and his description of the disease that bears his name have earned him an acknowledged place in nineteenth-century British medical history. Not as well known are Hodgkin's non-medical inte...

Phylogenetic origin of hepatitis B virus strains with precore C-1858 variant.

Mutations that prevent the expression of the hepatitis B e antigen frequently emerge in the immunoreactive phase of infection. The predominant mutation, the precore G-->A-1896 mutation, is restricted by the variability at position 1858 and is rare in strains with cytosine at nucleotide 1858. The C-1858 variant is characteristic of genotype A. It also occurs in genotypes C and F, but not in B, D...

Establishment of a new subfamily for Shenius anomalus (Shen, 1935) (Crustacea: Decapoda: Brachyura: Dotillidae)

Shenius anomalus (Shen, 1935) has previously been assigned to the Macrophthalmidae Dana, 1851, and the Camptandriidae Stimpson, 1858. However, a close examination of adult and first stage zoeal morphology confirms some previous suggestions that S. anomalus is actually an anomalous member of the Dotillidae Stimpson, 1858. However, because a number of distinctive features distinguish it from Doti...
